dc.description.abstract Supramolekulární chemie nabízí jedinečné možnosti, jak uspořádat molekuly do funkčních nadstruktur napodobujících principy biologických systémů. Jednu z rychle se rozvíjejících oblastí představuje vývoj umělých enzymů, jejichž aktivita je řízena změnami vnějších podmínek. Motivací této práce bylo vytvořit vícesložkový supramolekulární systém, jehož struktura i funkce by byly ovlivnitelné vnějšími podněty a který by mohl sloužit jako modelový nástroj pro biomimetickou katalýzu. Cílem disertační práce bylo navrhnout, syntetizovat a detailně charakterizovat nový heptatopický ligand schopný současně interagovat s několika typy makrocyklů (cyklodextriny, cucurbit[n]urily) i s kovovými ionty. Dalším cílem bylo ověřit, zda lze tyto interakce kombinovat za vzniku stabilních vícesložkových uspořádání s potenciální katalytickou aktivitou a zda je možné jejich funkci regulovat vnějšími podněty, například molekulárním signálem. V práci byly využity metody syntetické organické chemie, 1H NMR a UV-Vis spektroskopie. Ty nástroje umožnily jednak připravit používané komponenty a jednak popsat vazebné preference jednotlivých makrocyklů, jejich reorganizaci na ose ligandu, interakce s kovovými kationty, stabilitu vzniklých komplexů i jejich vzájemnou kompatibilitu. Výsledky ukázaly, že ligand selektivně váže různé makrocykly na specifická místa své struktury a kombinací těchto makrocyklů vznikají stabilní vícesložkové (pseudo)rotaxanové systémy. Byla rovněž jednoznačně potvrzena koordinace kovových iontů k bipyridinové jednotce ligandu. Navržené systémy byly následně testovány jako potenciální umělé enzymy, a to hydrolázy. Tyto testy byly nejúspěšnější při hydrolýze ethyl-formiátu (modelového substrátu), kde byla prokázána akcelerace průběhu reakce v přítomnosti navržených systémů. Tato práce představuje komplexní studii návrhu, organizace a funkce multikomponentních supramolekulárních systémů. Výsledky otevírají cestu k cílenému vývoji regulovatelných biomimetických katalyzátorů, jejichž aktivita by mohla být řízena molekulárními signály, změnou hostitele či reorganizací supramolekulární architektury.

dc.description.abstract-translated Supramolecular chemistry offers unique opportunities to organize molecules into functional architectures that mimic principles of biological systems. One of its rapidly developing areas is the formation of artificial enzymes whose activity can be regulated by changes of external conditions. The motivation of this work was to prepare a multicomponent supramolecular system whose structure and function could be modulated by external stimuli, serving as a model platform for biomimetic catalysis. The aim of this dissertation was to design, synthesize, and thoroughly characterize a new heptatopic ligand capable of simultaneously interacting with several classes of macrocyclic hosts (cyclodextrins and cucurbit[n]urils) as well as with metal ions. A further objective was to verify whether these interactions can be combined to form stable multicomponent assemblies with potential catalytic activity, and whether their function can be regulated by external stimuli, such as a molecular signal. Methods of synthetic organic chemistry, 1H NMR spectroscopy, and UV-Vis spectroscopy were employed to prepare the required components and to describe the binding preferences of the individual macrocycles, their reorganization along the ligand axis, their interactions with metal cations, and the stability and mutual compatibility of the resulting complexes. Results show that the ligand selectively binds different macrocycles at specific sites within its structure and that combinations of these hosts give rise to stable multicomponent (pseudo)rotaxane systems. Coordination of metal ions to the bipyridine unit of the ligand was also unambiguously confirmed. The constructed systems were subsequently evaluated as potential artificial enzymes, specifically hydrolases. The hydrolysis of ethyl formate (used as a model substrate) exhibited the most significant catalytic effect, with a clear acceleration of the reaction in the presence of the supramolecular assemblies. This work provides a comprehensive study of the design, organization, and function of multicomponent supramolecular systems. The results pave the way for the targeted development of controllable biomimetic catalysts whose activity may be regulated by molecular signals, host exchange, or reorganization of the supramolecular architecture.
